A general synthesis of dirhodium metallopeptides as MDM2 ligands.

چکیده
A synthesis of multifunctional dirhodium metallopeptide ligands for MDM2 is presented. An orthogonal protection scheme of palladium-catalyzed de-allylation on a metallopeptide substrate allows specific dirhodium incorporation in a complex peptide. Sequence effects on MDM2 binding are discussed.

We report a dirhodium paddlewheel compound which has an Rh2(5+) core enveloped by four guanidinate ligands and an axially coordinated bromine atom; Rh2(hpp)4Br crystallizes as a 1-D chain with a Rh-Rh distance of 2.430(3) è and a Rh-Rh bond order of 1.5 (hpp = the anion of 1,3,4,6,7,8-hexahydro-2H-pyrimido[1,2a]pyrimidine).

Complexes of novel alkyne-chelating tridentate ligands bound to a rhodium atom were isolated and characterised. The present alkyne-rhodium complex underwent dimerisation simply by heating to afford the unprecedented cyclobutadiene dirhodium complex. It is also found that the ligands at the trans positions influence the π-coordination of alkynes.
